چکیده

Gastric ulcer is caused by Helicobacter pylori which also can cause peptic and gastric cancer. The rise in the global prevalence of antibiotic resistance leads to a continuous decrease eradication rate H. infection. Recently, probiotic treatment, especially Lactobacillus reuteri supplementation has proven show inhibitory effects on able reduce antibiotic-associated side effects. Yet, efficacy treatment hampered lower cell mass production, mainly due low efficiency fermentation process. In this study, cultivation medium was optimized combining two different carbon-sources enhance multiplication. Cultivation media components were conventional statistical methods increase mass. optimization, Plackett-Burman design used screen significantly contributing factors their composition then using response surface methodology. contained (in g L-1): lactose, 30.0; glucose, 45.15; yeast extract, 33.0; K2HPO4, 2.0; CH3COONa, 3.0; C6H17N3O7, MgSO4, 0.5 MnSO4, 0.05. After culturing medium, biomass L. produced 4.12 L−1 compared 1.27 sole carbon-source MRS medium. addition, highly efficient production process developed batch bioreactor system. double-carbon source had improved fermentation.

Lactobacillus reuteri-specific immunoregulatory gene rsiR modulates histamine production and immunomodulation by Lactobacillus reuteri.

Human microbiome-derived strains of Lactobacillus reuteri potently suppress proinflammatory cytokines like human tumor necrosis factor (TNF) by converting the amino acid l-histidine to the biogenic amine histamine. Histamine suppresses mitogen-activated protein (MAP) kinase activation and cytokine production by signaling via histamine receptor type 2 (H2) on myeloid cells. Investigations of the...

Molecular characterisation and biomass and metabolite production of Lactobacillus reuteri LPB P01–001: A potential probiotic

Lactobacillus reuteri LPB P01-001 was isolated from the gastrointestinal tract of wild swine and was characterised by biochemical testing and sequencing of gene 16S rRNA. A simple and low-cost culture medium based on cane sugar (2.5% p/v) and yeast extract (1% p/v) was used in the production of this probiotic. The fermentative conditions were a) pH control at 6.5 and b) no pH control; both were...

Optimization of carbon source and glucose feeding strategy for improvement of L-isoleucine production by Escherichia coli

Fed-batch cultivations of L-isoleucine-producing Escherichia coli TRFP (SGr, α-ABAr, with a pTHR101 plasmid containing a thr operon and ilvA) were carried out on different carbon sources: glucose, sucrose, fructose, maltose and glycerol. The results indicated that sucrose was the best initial carbon source for L-isoleucine production and then sucrose concentration of 30 g·L-1 was determined in ...

Significantly enhanced biomass production of a novel bio-therapeutic strain Lactobacillus plantarum (AS-14) by developing low cost media cultivation strategy

BACKGROUND Probiotic bacteria are becoming an important tool for improving human health, controlling diseases and enhancing immune responses. The availability of a cost effective cultivation conditions has profound effect on the efficiency and role of probiotic bacteria. Therefore the current study was conducted with an objective to develop a low cost growth medium for enhancing the biomass pro...

Improvement of biomass yield and recombinant gene expression in Escherichia coli by using fructose as the primary carbon source.

The feasibility of substituting glucose with fructose as a carbon source in Escherichia coli fermentations was investigated. Glucose, the most commonly used sugar in bacterial cultivations, is well-known to pose a number of drawbacks; the most important of which is the Crabtree effect, which results in acidogenesis. Fructose, a glucose structural isomer, offers a reasonable alternative for gluc...
